I noticed that my Asus Prime B650M-A II motherboard is missing the screw thread for the 2280 M.2 slot. I've tried using an M.2 anchor, but I'm not sure if that's the right solution. I even attempted to remove the screw thread from the 2242 slot, but that didn't work either. Does anyone know how to resolve this issue? Thanks!

Have you checked the user manual? It should provide information on how standoffs should be set up for that motherboard. Sometimes manufacturers assume you have everything included, so it might just be missing.
It should definitely come with the motherboard. If you're still having trouble, consider reaching out to Asus support for help. They might be able to send you the missing part.
You might want to check out this mounting screw kit I found online. Asus has really dropped the ball by not including those extra standoffs anymore. It's frustrating when they save a few bucks and we have to deal with the hassle.

But be careful, this kit might not work for your specific board if there aren't any screw holes to use. It sounds like it needs to be pre-installed.
