I've read that Gsync performs best with Vsync set to 'On' in the Nvidia control panel, but it should be turned off within the game itself. Just make sure your monitor is Gsync capable—not just a 'Gsync compatible' display, since settings can differ a lot.
I prefer leaving Vsync at its default setting in the Nvidia app and turning on Reflex for all my games. I found that this naturally caps my framerate to a bit below my monitor's refresh rate, which helps in maintaining smooth gameplay.
