My situation is that I have a Fire Stick connected to my monitor's HDMI port, and I'd like the audio to route through my PC to take advantage of my better speakers. I know I could just use the monitor's audio out jack to hook up to the PC's Line-In, but would it be possible to achieve this directly through the DisplayPort connection? My setup would look like this: Fire Stick -> Monitor HDMI Port -> Monitor Audio Out -> PC (via DisplayPort cable) -> PC Speakers.

I get that you're looking for a cleaner setup, especially to control volume from your PC. But since your PC speakers only take one input, you might just have to stick with the monitor’s aux out for now. It would be awesome if DisplayPort added this feature in future updates, but since using a Fire Stick with PC monitors isn’t super common, it might not be on their priority list.
Unfortunately, DisplayPort doesn’t support sending audio back to your PC. It only transmits audio when it’s outputting from the PC. HDMI is capable of two-way audio, but most computers don’t have that feature either. If your speakers are standard analog ones, the simplest option is to connect them straight to the monitor's analog output instead of routing through the PC. It’s just more straightforward that way!
