I'm currently building a PC and I've noticed that my motherboard has 10 mounting holes while my case only has 9 standoffs. I'm wondering if it's alright to leave one of the holes empty, or if I might be missing something important in the setup.

Double-check your case package; there might be spare standoffs included. If you’re missing one, it won’t be a big issue as long as the critical areas have support, especially around where the power supply connects.
If you’ve got 9 standoffs, you should be good! Most motherboards only need a handful to stay secure, so 90% coverage is usually enough. Just make sure you have standoffs where the power connectors are for extra support.
You're totally fine leaving that hole empty; having 9 screws will securely hold your motherboard in place. It's more about the stability from the available standoffs than hitting every single hole.
No big deal if one hole is missing a standoff. Many new cases are designed with that in mind. Some boards have a hole for compatibility with older cases that had more standoffs, but if you don't have one there, just screw into the ones you do have without worry.
