Why am I getting a ‘file too large’ error when copying to my USB drive?

I'm trying to transfer a 4GB .mkv file to a 32GB USB thumb drive, but I keep getting an error saying the file is too large for the destination. I've checked several empty and nearly empty flash drives but keep encountering this issue. Can anyone explain what the real problem is here?

The most common issue here is likely that your flash drive is formatted as FAT32. Right-click on your drive in Windows Explorer, choose 'Format...', and you can reformat it to NTFS. Just keep in mind that this will delete everything on the drive!
