I'm setting up an older DDR3 computer for my family and I have both an HDD and an SSD, each with a clean Windows 10 install. They work perfectly fine when tested on another computer. However, on this particular machine, it only boots from the HDD when the SSD is also connected. If I try to boot with just the HDD, I get a 'non-system disk/disk error' message. What could be causing this issue?

It sounds like the boot loader might be located on the SSD instead of the HDD. You may need to try rebuilding the bootloader on the HDD to fix this issue.
You can't just swap OS drives between different PCs as they might not be compatible. I suggest you try installing a fresh copy of Windows on the HDD. Just note that you'll need to make sure the computer can boot from your installation media first.
