I'm upgrading my motherboard from a micro ATX to an ATX size. I'm curious if the new motherboard will come with standoffs, or do I need to take the ones off my current setup? I'm not sure how to remove them either.

Yeah, in my experience, you'll find that MOBO standoffs usually come with the case. If you don't have enough, you can purchase a packet of case screws on Amazon, which includes standoffs along with other screws you might need for your build.
Generally, standoffs are included with the case and not with the motherboard itself. That means you'll need to use the ones that are already in your case. If you need replacements, they're easy to find at computer hardware stores.
Removing standoffs is pretty straightforward. They usually just twist out like screws. If they're stuck, you can use pliers to help turn them. Make sure to check the positioning for the ATX; it should have slots for the standoffs to match up just fine!

If you have the tools, a small socket wrench can make it easier to get them out. But, if not, just go for the pliers; just remember lefty-loosey!
