I recently relocated the Cities: Skylines 2 game from my C drive to my E drive, but after doing so, all 500GB of my E drive suddenly appeared on my desktop. I've tried restoring my computer to a previous point, but that hasn't worked. Is there anything else I can try to fix this?

When you moved the game, did you create shortcuts instead of moving the files? Make sure you verify if the actual files were shifted to the desktop or if it just created shortcuts.
Could it be that you accidentally set your E drive as a desktop library? That sometimes happens when you relocate files. It's worth checking your libraries settings!
What method did you use to move the game? If the game files are appearing directly on your desktop, it could be that they got linked rather than moved. More details on how you performed the move would help troubleshoot this better.
Have you checked if the files are just shortcuts on your desktop? It's possible that nothing was actually moved, and your E drive might still be intact. You can look at the free space on both drives to confirm whether they are actually connected. A tool like WizTree can help visualize your storage for clarity.
