I have a graphics card with only one HDMI port, and I'm looking to connect a second monitor, which is an XP-Pen tablet. Will an HDMI to DisplayPort adapter work for this setup?

Just make sure you're using the right type of adapter. If your tablet has HDMI and the GPU has DisplayPort, you'll need a DisplayPort to HDMI adapter, not the other way around.
You're right that a simple DP to HDMI cable works well for this. Just avoid the HDMI to DP adapters as they are different.
I've tried using an HDMI to DisplayPort adapter, and it worked for me! But I think there's a limit to the resolution you can get, so keep that in mind.
If your graphics card only has one output, it might be getting a bit old. You could also explore using a USB-C video output if your setup allows, but it might not be supported.
