My main concern is about clearance with the AIO cooler and whether everything will fit nicely, especially with the vertical GPU mount and using riser cables. I found some low-profile RAM from a Microcenter bundle that works for now, but my plan is to upgrade to 64GB later. If your main use cases are for the 5080 running LSFG and using both GPUs for RIFE + Topaz processes, the X4 might not be a dealbreaker for you. Those tasks shouldn't be bottlenecked. I was considering the MSI ACE for a faster second slot myself, but your choice does seem solid overall. Have you explored any other motherboard options?
The warning you got isn't about clearance but rather the PCIe lanes. The Asus ROG STRIX X870E-E is a great motherboard, but just keep in mind that its second PCIe slot only runs at X4 mode instead of X16, which could limit your second GPU's performance depending on what you're using it for.
