I'm considering using a single 4TB 990 PRO NVME drive for my PC. Is it practical to keep Windows, all my games, and everything else on just this one drive?

Yes, it's totally fine to go with one big NVME drive. You don't need to create separate partitions unless you plan to reinstall Windows frequently. Just keep everything on one partition, so you don't have to deal with space management issues later on.
